  • 摘要:   目的  利用癌症基因组图谱(the cancer genome atlas, TCGA)数据库分析胃腺癌组织中赖氨酸羟化酶3(procollagen-lysine, 2-oxoglutarate 5-dioxygenase, PLOD3)基因表达及其潜在临床价值。  方法  使用UALCAN数据库(http://ualcan.path.uab.edu/)分析TCGA中PLOD3基因在胃腺癌及癌旁组织间表达差异;利用Kaplan-Meier模型分析PLOD3基因表达与胃腺癌患者预后相关性;利用LinkedOmics数据库筛查PLOD3相关基因;应用免疫组织化学验证胃腺癌与癌旁正常胃黏膜组织PLOD3来源与分布差异。  结果  数据库资料表明,胃腺癌组织PLOD3基因表达高于正常胃组织(P<0.001);伴有p53基因突变的胃腺癌患者PLOD3基因表达高于无p53基因突变患者(P<0.001);PLOD3基因表达与胃腺癌患者生存期相关(HR=1.360,95% CI:1.150~1.610,P < 0.001);PLOD3基因表达与AP1S1、LRWD1和SLC12A9等基因表达呈正相关(均有P<0.001),与CGGBP1、ZNF561和MIER1等基因表达呈负相关(均有P<0.001);免疫组化染色结果进一步证实胃腺癌组织PLOD3蛋白表达高于癌旁正常胃黏膜组织(P=0.031)。  结论  PLOD3可能成为评价胃腺癌预后以及胃腺癌靶向治疗的有效新靶点。
